E. coli/Quantitube® Medium This medium indicates and identifies the presence of E. coli in a test sample. The E. coli form small, dark teal green colonies with a teal halo, while other members of the Enterobacteriaceae grow as dark red colonies without a halo. For optimum results, the medium should be incubated at a temperature of 35 C, and results counted at 24-48 hrs. The teal green color (from the chromogenic substrate used) is specific for the enzyme glucuronidase, which is virtually exclusively found in E. coli among the coliforms.

This is a High-Sensitivity 5 ml water or liquid/soft food E. coli test.

Remove the E. coli Quantitube(® Strip from the Quantitube® . ID Label may be removed and reapplied several times; almost any pen will write on the label.

Use the Graduated Cup to measure any liquid or soft food. Orange Juice was used for this demonstration. Use the pipette place 5ml of any liquid or wet food into the graduated cup. Dried foods and grains may be washed or allowed to soften and devolve in sterile or cooled boiled water.

Pour the liquid into the Quantitube®Medium. Reseal the media and gently invert 3 times.

Pour the liquid Medium into the Quantitube®.

Insert E. coli Quantitube® Strip and cap. Allow 1 hour for E. coli Quantitube® Medium to solidify; store upright in incubator box.

Instructions

This is a High-Sensitivity 5 ml water or liquid/soft food E. coli test.

The stand holes for the Quantitube(® are under the label of the Box. Using a Quantitube®, push through the label to open the stand hole; there are 5 holes.

Number the Quantitubes® you are going to use for your sample and fill in the Chain of Custody form. The tape can be removed for counting and inspection and then reapplied.

Sampling and measurements: The E. coli Quantitube® Test requires a 5ml liquid sample. For your convince, there are two way that you can measure your sample: the graduated Quantitube®) or the graduated cups.

Field water samples may be collected in the capable graduated Quantitube®.

Samples may be measured in the graduated Quantitube(®) or the graduated cups included in your kit.

5 ml of any liquid may be added to the Quantitube®. One bottle of E. coli Quantitube(® Medium should then be added to the Quantitube(TM) . The cap should be pressed on firmly and the Quantitube(® inverted three times. Do not shake; gently invert the Quantitube(® three times.

Remove the cap of the Quantitube(® and insert the a Quantitube® Strip narrow end first into the Quantitube®.. Reseal the Quantitube(TM) and insert into the box stand for incubation.

Using the graduated cup may be easier for some people. Squeeze the cup to form a pour spout. Measure your sample into the cup. Pour the sample into the E. coli Quantitube(® Medium. Reseal the bottle. Gently invert the Quantitube® Medium three times. Remove the cap from a Quantitube(TM) . Insert the a Quantitube(® into the Box 1 stand. Now pour the contents of the E. coli Quantitube(TM) Medium with the 5 ml sample into the Quantitube(®. Insert the a Quantitube(® Strip narrow end first into the Quantitube(®. Reseal the Quantitube(® .

 
Why This Test Works
